Research summary
Analysis of a responsible gene, Necdin of neurodevelopmental disorder, Prader-Willi syndrome and the functions of the related genes Necdin is an intracellular protein with approximately 45kDa that is expressed and induced in the brain, particularly, in the hypothalamus just after the generation of neurons (about 10 days later when the neurons are generated in a mouse) and human Nectin genes exist in a chromosome 15q11-12 which is a Prader-Willi syndrome-deleted region and is considered as a causative gene of the syndrome. Main symptoms of Prader-Willi syndrome are short stature due to hypothalamic hormone deficient secretion, abnormal sex organ growth, hypotonia of muscles, breathing disorder, mild intellectual impairment, impaired peripheral sensation, adiposeness and so on. Similar symptoms are recognized in a Necdin-deleted mouse, and the symptoms such as decrease of hypothalamic hormone-producing cells, death shortly after birth due to breathing disorder, decrease of dorsal root ganglion neurons, abnormal neuron dendrite extension in brain, developmental anomaly of GABAergic cortical interneurons and adiposeness due to high fat diet are reported. It is reported that breathing disorder is also caused by abnormal breathing center neurons. It is implied that Nectin is involved in the regulation of cell death of nerve cells and cell differentiation. It is reported that it is involved not only in nerve cells but also in differentiation of muscle cells and adipose cells. Necdin belongs to the MAGE family and more than 30 kinds have been reported so far and it is implied that the protein group out of them that expresses in the differentiated cells called TypeII has redundant functions. A MAGE Homology domain consisting of approximately 200 amino acids exists in common for the MAGE family and approximately 50% of homology to each other is recognized. The MAGE genes are stored in a wide range of species from plants to higher organisms, it is the only one kind out of low organisms. In recent years, it was made clear that a Nse3 out of Smc5-Smc6 complexes is a single MADE gene. It is reported that the Smc5-Smc6 complex is involved in DNA repair, but its functions are not yet known. Nse1, Nse3 and Nse4 form a sub-complex in the Smc5-Smc6 complex. Nse4 belongs to the EID family and it is reported that it is involved in the cell differentiation by restraining transcriptive activity. In addition, although Nse1 has a RING finger structure similar to Ubiquitin ligase, the functions are not yet known. Based on these facts, I will perform research to analyze Nse1, Nse3, Nse4 and the functions of their binding protein, and clarify the ripening mechanism of the cell differentiation through the basic function analysis using simple organisms such as cellular slime mold. Furthermore, I would like to clarify the pathogenic mechanism of Prader-Willi syndrome through these function analyses.
